Antibody-forming cells (AFC) directed toward antigens on the sheep red blood cell have been localized to the sacculus rotundus of the normal unimmunized adult rabbit. These cells are either potential AFC or direct precursors of the AFC. The normal rabbit sacculus does not possess antigen-reactive cells (ARC) directed toward this or any other antigen. Only the rabbit bone marrow possesses the ARC. The significance of this finding with respect to accepted criteria for the definition of the primary immune response, both in vivo and in vitro is discussed.
