In Results, the authors wish to provide clarification of the experiments using the arts-1(−/−) cell lines in Figures 3 and 4.
Although the cloned arts-1(−/−) cell lines utilized in this study did not express either ARTS-1 protein or mRNA, and arts-1 exons 5 and 6 could not be amplified by PCR of genomic DNA, subsequent experiments have revealed detectable ARTS-1 protein expression in these cell lines. Therefore, the arts-1(−/−) cell lines are not clonal populations of cells homozygous for the targeted deletion of arts-1 exons 5 and 6. This was confirmed by PCR of genomic DNA using the arts-1 exon 5/6 primer pair. As shown below in Figure 8, a 1328 PCR product was generated from the wild-type NCI-H292 cells, as well as from the three arts-1(−/−) cell lines (#1–#3), which demonstrates the presence of arts-1 exons 5 and 6. Since the arts-1(−/−) cells that were used for the experiments presented in this paper expressed neither ARTS-1 protein nor mRNA at the time the experiments were performed, we believe that the conclusions regarding the role of ARTS-1 in IL-1R shedding are valid.
