Information has been presented regarding two methods for testing the passive mouse protective power of sera from subjects receiving Shigella vaccines. These two methods are: the Constant Serum Passive Mouse-Protective Test and the Constant Organisms Passive Mouse-Protective Test.

We prefer the “constant organisms” method since it permits comparison of protective volumes of sera and the calculation of the increase in protective power of serum obtained after vaccination as compared to serum from the same subject before vaccination. The “constant organisms” method in our hands requires fewer mice and less serum.
